Background: Synthesized aminocoumarins are heterocyclic compounds possessing potential for the treatment of insulin-dependent diabetes mellitus with unexplored anti-glycative action. Results: In this study 4-aminocoumarin derivatives (4-ACDs) were evaluated in vitro for antiglycation (AG) activities by using the human serum albumin (HSA)/glucose system, for 8 weeks of incubation. The glycation and conformational alteration of HSA in the presence of the tested compounds were evaluated by Congo red assay, fluorescence and circular dichroism spectroscopy. The antioxidant (AO) capacity were also tested by four different assays including: DPPH (2,2'-diphenyl-1-picrylhydrazyl radical), ABTS (2,2-azinobis (3-ethylbenzothiazoline-6-sulphonate) diammonium salt),... |

Background: Interleukin 8 is a chemokine that is produced by several types of cells, like macrophages and has chemotactic activity in particular on neutrophils, playing a key role during the inflammatory process. It has been demonstrated at the molecular level that this molecule is present and conserved in several vertebrate groups, pointing its importance. Analysis of the amino acid sequence of IL-8, projected from cDNA of Salmo salar, presents homology with the sequences of mammals, poultry and lamprey, indicating the presence of a homologous molecule in higher fish. However, there is no information at protein level, which allows characterizing the regulatory role of this molecule during the immune response in fish. Results: In this work, we designed and... |

Background Polycosanols derived from plant species have traditionally been used in medicine as antiproliferative agents for treating various viruses (primarily the herpes simplex virus). However, few studies have studied their effects on hyperproliferative cell lines. In this work, the antiproliferative capacity of polycosanols from tall-oil pitch, obtained from black liquor soaps in the kraft pulping process of cellulose (specifically from Pinus radiata, Pinus taede, and Eucalyptus globulus), was evaluated on CHO-K1 and CRL-1974 human melanoma cell lines. Results The proliferative capacities and cell viabilities were measured for 72 and 140 h, respectively. Treatment with docosanol produced differential effects on the CHO-K1 and human melanoma cells and... |

Background: Bioleaching of minerals is a process that allows the treatment of insoluble sulphides and insoluble oxides via hydrometallurgy, as opposed to the more traditional technology of pyrometallurgy. Bioleaching presents several technological, environmental and economic advantages as compared with pyrometallurgy, as well as some limitations. Results: In this work a general panorama of copper mining and biomining, the influence of the most relevant operating variables and their present and future situation in Chile are presented and discussed. Conclusions: Today, Chile plays an important role in the development and commercial application of bioleaching to copper ores. This background information allows predicting a brilliant future for this technology... |

Single Strand-Preferring Nucleases (SSPNs) have been implicated in the triggering and progress of cell death pathways, including senescence in higher plants, though the biological roles of such enzymes are still obscure. In the present study, heterotrophic cell suspension cultures of Oryza sativa L. (rice) cv Taipei 309 were used to investigate changes in Single Strand-Preferring Nuclease activity associated with cell death in vitro. An acid nuclease activity (pH 5.5) was found which was strongly stimulated in the presence of 10 mM Zn2+ and inhibited by 10 mM EDTA and EGTA. An increase in SSPN activity was concomitant with a loss of cell viability, total protein and the onset of stationary phase of growth in the cell cultures. Using DNA-SDS-PAGE two major... |

Changes in the DNA content and organisation of senescing rice cell cultures (Taipei 309) were studied, using PCR and Southern blot analyses. A mitochondrial gene (coxII), a plastid gene (psaA) and a nuclear DNA maker (RG64) were analysed. The amplification of mitochondrial (mt), plastid and nuclear DNA produced the expected fragments, indicating that there were still some intact organelles and nuclei in the senescing rice cells. However, in plastid and nuclear DNA, changes in the number and size of the PCR products were observed. Southern blot analysis revealed that mt and nuclear DNA contents declined, but plastid DNA content remained relatively the same throughout the senescence. |

Biotechnology, in its voyage from ancient times into the expanding frontier areas of today's world of scientific research, has revealed itself as the gene of diplomacy and international cooperation in the relationships between nations (<a href="#16">DaSilva, 2002</a>). Economic and biotechnological considerations, strong elements in themselves, drive policy implementation that draws upon the support and sustenance of the public's logic and understanding of science obtained through the avenues of art and culture. More and more scientists are teaming up with artists to harness the world of microbes and biotechnology in the pursuit of one's natural well-being, of the conservation of environmental heritage, and of human comradeship and welfare. Background: Glycine oxidase (GO), a type of D-amino acid oxidase, is of biotechnological interest for its potential in several fields. In our previous study, we have characterized a new glycine oxidase (BceGO) from Bacillus cereus HYC-7. Here, a variant of N336K with increased the affinity against all the tested substrate was obtained by screening a random mutant library of BceGO. It is observed that the residue N336 is invariable between its homogeneous enzymes. This work was aimed to explore the role of the residue N336 in glycine oxidase by site-directed mutagenesis, kinetic assay, structure modeling and substrate docking. Results: The results showed that the affinity of N336H, N336K and N336R increased gradually toward all the substrates, with increase... |

Background: Inulinases have been extracted and characterized from inulin-storing tissues; however, production of microbial inulinases have recently draw much attention as they offer several industrial advantages. Many microorganisms, including filamentous fungi, yeast and bacteria have been claimed as inulinase producers. These hydrolases are usually inducible and their exo-acting forms may hydrolyze fructose polymers (inulin) and oligosaccharides such as sucrose and raffinose. Fungal inulinase extracts are often produced as stable mixture of highly active fructanhydrolases. From a practical prospective, the best known inulinases to date are those produced by species of Penicillium, Aspergillus and Kluyveromyces. Results: The production of extracellular... |

Sesame (Sesamum indicum L.) is one of the oldest oil crops and is widely cultivated in Asia and Africa. To determine the level of genetic diversity in relation to geographical origins and morphological characteristics, a total of 96 accessions have been collected from different parts of the world and were analyzed using AFLP techniques. Twenty-one primer pairs generated a total of 445 bands and among them 157 (35%) were polymorphic. Using UPGMA clustering analysis method based on the similarity coefficient, accessions were separated into two major groups. The first group mostly consists of Eastern Asian origin and another group consists of South Asian origin. Sub-clusters separated the accessions and form distinct diversity among groups. Considering the... |

In Venezuela, pedigree analyses indicate that the rice varieties currently under cultivation are closely related. Effective breeding programs, based on knowledge of the genetic diversity of cultivars, are needed to broaden the genetic bases of rice germplasm in the country. In this study, we used a set of 48 simple-sequence-repeat (SSR) markers to assess the genetic diversity of 11 Venezuelan rice cultivars, released by the National Rice Breeding Program between 1978 and 2007. A total of 203 alleles were detected, the number of alleles (NA) per marker ranged from 2 to 9, with an average of 4.23. The average genic diversity (H) over all SSR loci for the 18 genotypes was 0.524, ranging from 0.105 to 0.815. Positive correlations were found between H at each... |
